In that case, turn on Lossless mode. Lossless WebP keeps every pixel and the transparent background exactly as in the PNG, so nothing is lost before the printer sees it; lossy WebP, even at the default quality of 90, discards detail you might notice on paper.
Transparent areas behave differently in print: they usually receive no ink and show the paper color. If the design needs a white or colored panel behind it, fill it before printing, for example with PNG to JPG and a chosen fill color or by placing it on a PDF page. Pixel count sets print size as usual: at 300 pixels per inch, a 3000-pixel-wide design prints 10 inches wide.
